To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Reducing sugars assays are quantitative, sensitive and almost usable with all kind of polysaccharide but some compounds may interfere with them. When polysaccharide is... |

Estimates of carbohydrate assimilation within the mesozooplankton compartment are a fundamental concern in oceanography because they give a general expression of the biologically-regulated incorporation of metabolic fuel produced from inorganic nutrients by phytoplankton in the sea. A new tool is proposed for the assessment of such processes. It is based on the measurement of the activity of an enzyme, pyruvate kinase (PK), on samples of the whole mesozooplankton community. Here, the results are presented of an early attempt at such an implementation in the Bay of Biscay (north-east Atlantic) in springtime. A main area situated in front of an estuary (Gironde) has been extensively studied-the same sampling grid was covered on three occasions (two 20 day... |

In order to develop innovative methods for the production of bioactive extracts with potential health benefits, enzyme-assisted extraction has been applied for the bioconversion of Ulva armoricana (Ulvales, Ulvophyceae). The extraction yields, chemical composition, the antioxidant and antiviral activities were determined to assess the efficiency of six commercial enzymatic preparations. Endo-protease treatments significantly increased the extraction yields compared to the control. The organic matter, neutral sugar and protein contents were increased in all extracts compared to an extraction with water, up to 2.0-fold, 2.7-fold and 1.75-fold, respectively. The samples prepared by a multiple-mix of glycosyl-hydrolases and an exo-β-1,3(4)-glucanase revealed... |

Phosphorus (P) limitation of phytoplankton growth is known to affect the accumulation and release of carbohydrates (CHO) by micro-algae. However, relatively little is known about the fate of algal exudates, notably their bacterial degradation. The CHO chemical characterization is also not exhaustive, especially in ‘functional’ pools relevant for phytoplankton physiology (particulate reserve [R] or structural [S] CHO) and for bacterial degradation (dissolved mono- [MDCHO] and polysaccharides [P-DCHO]). In this study, we investigated how P depletion and repletion affect the CHO composition in diatom Thalassiosira weissflogii cultures, and the shortterm response of free and diatom-attached bacteria in terms of abundance and potential βglucosidase activity... |

Enteric pathogens cause considerable public health concerns worldwide including tropical regions. Here, we review the roles of carbohydrates in the infection strategies of various enteric pathogens including viruses, bacteria and protozoa, which infect the epithelial lining of the human and animal intestine. At host cell entry, enteric viruses, including norovirus, recognize mainly histo-blood group antigens. At the initial step of bacterial infections, carbohydrates also function as receptors for attachment. Here, we describe the function of carbohydrates in infection by Salmonella enterica and several bacterial species that produce a variety of fimbrial adhesions. During invasion by enteropathogenic protozoa, apicomplexan parasites utilize sialic acids... |

The objective of this work was to evaluate the effect of inclusion of dietary glycerol in replacement to starch on the growth and energy metabolism of Nile tilapia juveniles. The experiment was carried out in a completely randomized design with four treatments (0, 5, 10, and 15% purified glycerol) and six replicates. Pelleted, isonitrogenous, and isocaloric diets were provided for 60 days. Growth performance parameters and muscle glucose and protein concentrations were not affected by dietary glycerol levels. The treatment with 15% glycerol presented higher levels of muscle and liver triglycerides. A quadratic effect of treatments on muscle and liver triglyceride concentrations was observed. The treatment with 0% glycerol presented higher hepatic glucose... |

The guava (Psidium guajava L.) cv. Paluma has been cultivated in São Francisco Valley, Northeastern of Brazil, for in natura consumption and processing purposes. In spite of its importance, there are few scientific knowledge regarding guava physiology, nutrition, irrigation and fertigation. The objective of this work was to evaluate the effect of weather conditions and different concentrations of N and K applied by fertigation in foliar contents of reducing sugars, total soluble sugars, starch, sucrose, amino acids, and proteins. The field experiment was carried out at Bebedouro Experimental Field and the biochemical evaluations at the Laboratory of Seed and Plant Physiology, both located at Embrapa Semi-Árido, Petrolina-PE. The doses of 200 g N and 100 g... |

Carbohydrates can provide up to 80% of the dry matter in animal diets, yet their specific evaluation for research and diet formulation is only now becoming a focus in the animal sciences. Partitioning of dietary carbohydrates for nutritional purposes should reflect differences in digestion and fermentation characteristics and effects on animal performance. Key challenges to designating nutritionally important carbohydrate fractions include classifying the carbohydrates in terms of nutritional characteristics, and selecting analytical methods that describe the desired fraction. The relative lack of information on digestion characteristics of various carbohydrates and their interactions with other fractions in diets means that fractions will not soon be... |

Most of the plant regeneration processes in citrus, through tissue culture, involve indirect somatic embryogenesis. The optimization of these processes is important for the development of in vitro plant improvement and micropropagation studies. Studies to evaluate the effect of different carbohydrates in somatic embryogenesis were conducted using calli from 'Ponkan' mandarin (Citrus reticulata, Blanco), 'Cravo' mandarin (C. reticulata), 'Itaboraí' sweet orange (C. sinensis L. Osbeck.), 'Valencia' sweet orange (C. sinensis) and 'Kinnow' mandarin (C. nobilis Loureiro x C. deliciosa Tenore). The culture medium used was MT supplemented with sucrose, galactose, glucose, maltose or lactose with the following concentrations of 18, 37, 75, 110, and 150 mM. The... |

Research about the use of natural light associated to changes in sucrose levels demonstrated potential in promoting in vitro hardiness of tropical climate species, as well as reducing production costs. However, little is known about physiological and structural changes that happen in the process. This study evaluated the physiological and anatomic performance, and ex vitro survival of micropropagated banana plants in response to cultivation conditions, in the stage of in vitro rooting. Shoots of the 'Caipira' cultivar were cultivated in MS medium, supplemented with 1 mg L-1 NAA and 6 g L-1 agar, in which the following treatments were applied: two sucrose concentrations (15 g L-1 or 30 g L-1) and two cultivation conditions (Natural light - greenhouse and... |

Aim of this research was to investigate physical characters and some chemical characters of northeast local rice grain of Thailand. Physical characters were studied in term of weight, width and size of seed and dehusked seed including elongation rate and water absorption rate of 15 rice cultivars (cv.). Chemical test were checked by protein content, amylose content, lipid content, carbohydrate content, vitamin B1, vitamin B2, vitamin B3 and vitamin B6 content. The result showed that the longest seed of 15 cv. was Jowluang, 10.82 and 7.81 mm. in seed and dehusked seed, respectively. In contrast, cv. Kowjowhangmuyai (GS no: 554) showed the shortest seed and dehusked seed was 7.68 and 5.29 mm, respectively. The largest seed and dehusked seed were found in... |
